I am trying to do a SELEX to find DNA aptamers against few selected hydrophobic peptide targets of length 13-17 mers but not yet able to obtain successful candidates. I tried with different types of DNA aptamer library, but not able to get proper bands in gel to confirm the presence of candidates. Can anyone help with suggesstions, ideas, etc.?
During selection step, did you immobilized your peptide targets on any substrates like beads or resins?
For my peptide target, it contains 33 mers but because the length of the target is too short and molecular diameter is not bulky, I was advised to add a spacer in between 33-mer peptide and the beads.
Yes Suttinee, I had tried different beads. I immobilised my peptides into Dynabeads Myone carbolxylic acid, but I didnot have any spacer. Yea my target peptides are also shorter one with 13-17 mers.
