I am currently working on developing a bioluminescent mouse model for investigating the anti-leukemic activity of our test compounds. For that purpose I wish to conduct the lentiviral transfection of firefly luciferase into human leukemia cell line MV4-11. As I am going to do this for the first time I have no background as to which vectors encoding for the gene of interest should be used. Also which of the kits available commercially for this purpose are better. I have gone through protocols from Promega, Thermo-Fischer, Takara, Qiagen to name a few. Looking for some answers and suggestions.

Do you really need lentiviral transfection? If you are trying to get firefly luciferase into a cell line there are other transfection techniques that could be simpler to execute (electroporation for example: https://altogen.com/product/k562-electroporation-kit-aml-leukemia-cells-ccl243/). MV4-11 isn't a common cell line, and it would do you good to research lentiviral/transfection methods that are used in context of other AML cell lines. Electroporation is usually used in suspension cell lines, though you should be able to reach high efficiencies with other methods as well.
I see you're in South Australia, as far as I know the University of Adelaide still has a viral vector core as part of the Robinson Research Institute (I used to run it). They should still sell stock viruses by the ul, I suggest that you purchase a small amount of GFP viruses with several different promoters and use them to see if your cell line can even be infected with a lentivirus. Unless you see good transduction, it's probably better to look at alternative methods as Ted has suggested
