I am currently working on primary MSCs derived from bone marrow of C57/BL6. I am able to grow them on PCL matrix but facing problems dissociating them for further passage. is it possible to dossociate MSC from PCL scaffold for further passaging or should i look into other alternatives?

Thank you for sharing the link. The given article describes the use of Collagenase IV to dissociate cells from a dynamic 3D culture system. However, I am currently using a static 3D culture system. I am not sure what would be the outcome of using collagenase IV on static 3D system. I would be grateful If you could share your thoughts on this.
I am running almost the exact same protocol tomorrow morning using trypsin as our dissociating reagent (mainly because that's what always works for passaging in flasks). I'll let you know how the results turn out.
I too have wondered the best procedure. I culture cells on PLGA/PCL and have tried trypsin with poor yield. Please advise on advice

Even we have opted to grow our cells on hydrogels for now. Thanks for the input. However Dr Marks mentioned he had some success in getting cells off 100%PCL using trypsin, albeit low yield. Lets look up to him for his future results.
