Hi, I don't work with cherries but I am extracting DNA with the DNA Extraction Kit from MasterPure (Epicentre). I am working with insects and for me this kit work very well! Cheers, Nadine

The quality of DNA is affected by the polyphenol and essential oil content of the material you use (bud). I m not sure if the kits you use has the provision for phenol extraction. Otherwise, phenol extraction step would help you in obtaining quality DNA...

I am extracting DNA from Variety of Plants including apple. But high phenol content reduces the yield content and quality. I am using CTAB method for DNA Extraction. The buffer contains 1% of PVP K-30; 2% CTAB and 0.5 % of mercepethanol. you can try this the NaCl concentration is 1.4M and TrisHCl and EDTA. Try this method surely you will get good quality of DNA

i worked with pomegranate (with almost 10 % phenols in it)and used Genei Kit , also used CTAB but without PVP purification. By both got good DNA. I think its a stage at which u take the samlpe. and of course very fine crushing. For me very young leaves which have emerged 1-2 days before ,did not worked.
