Hi. On general I am using Trizol reagent for total RNA extraction and on general it is working quite well, with very good RNA quality and yield, and almost no DNA contamination. Often before when i use CTAB method (it is good one) I have sometimes a lot DNA contamination and I do not like to treat my RNA with DNAseI.
yes, you can try TRizol also. I extracted RNA from root and leaf of rice plant using TRIzol (Invitrogen, Carlsbad, USA) following the manufacturer’s instructions and this RNA was subsequently used for first strand cDNA synthesis with the aid of Superscript ΙΙ (Invitrogen) using oligo dT primers.
The TRIzol (Invitrogen, Carlsbad, USA) was found good for isolation of sufficient amount of RNA.

If you can afford it buy a kit for RNA extraction, a kit for DNAase treatment and a kit for cDNA synthesis and follow the instructions. The latter two kits are more or less necessary and you can only perform RNA extraction without a kit. There are also some tricks that you can use during the process that me (and I believe anybody in the forum) can discuss when you need to.
