@ Sezer Okay. Thank you for your answer and article. Any Primers are required for C-DNA synthesis ? Means only dNTPs and RT Enzyme can do it or its required a set of primers ? Stem loops primers required once we have DNA, for amplification.
As Sezer has written stem-loop primers are good to quantify miRNAs. But if you have more samples (say 3 different time points) and you want to validate 5 miRNAs, you will have to synthesize 15cDNAs (5*3). In such case it is better to use, a kit such as miRNA synthesis kit from Clontech. I have used this kit and the results are really good (It saved me a lot of time as I had 7 time points in my experiment).
But if you are dealing with 1/2 samples and few miRNAs, go with stem loop primers. Hope this helps.
