I have conducted a CRISPR knockout in SHSY5Y cells and have carried out a few differentiations on them. We do an 8 day protocol using 10uM retinoic acid and 1% FBS which is fairly standard across many labs.
However, we see a large amount of epithelial type cells. We are interested in schizophrenia, so naturally are looking at neuronal morphology. As such, the increased amount of the epithelial cells can impede this (they are seen in both WT and KO).
I have been looking for previously published work to help explain this, but I haven't been able to come across anything. The serum starvation supposedly should take care of them. Some of the cultures are a bit older which may explain it?
