Has anybody used One Shot® OmniMAX™ 2 T1R Chemically Competent E. coli from Invitrogen? I could not get the result of my cloning with those, while same plasmid was successfully cloned in NEB 5 alpha cells.

I used both of them for gateway (LR) and classic clonning. I have no problem with them..
What kind of cloning you are making?
i have used them for both BP and LR cloning, but could not get the result.
have you checked the transformation efficiency of these competent cells, probably it has been lost.
I used DH5alpha for BP cloning and omnimax and DH5 for LR. I never had problems. For classic cloning I use only DH5 alpha, and don't forget to make a positive control that can help you to know if your bacterias are still efficients.
Hi céline, as far as i understand you use for your positive, negative and your specific BP reactions DH5alpha competent cells instead of the recommended One Shot® OmniMAX™ 2-T1R Chemically Competent E. coli, is that correct? For the positive control is suggested to use DH5alpha, but i was wondering whether i always have to use One Shot Omni Max competent cells for negative and specific BP reactions? DId you have success in cloning larger inserts ( apprx 9000 bp) with Gateway system? If yes, can you share some tips with me? Thank you in advance for your reply! best wishes
