An established in-house protocol has so far proven unsuccessful to stain tumor samples for macrophages using F4/80 (CI:A3-1) : sc-59171. It would be highly appreciated if someone can share IHC protocol using this antibody, particularly the antigen retrieval method. Flow analysis has shown presence of macrophage populations in the tumor tissue. General suggestions from experience with other antibodies are also welcome. Thank you.
Sc-59171 seems like a SantaCruz antibody. I didn't find any specific protocol about this product, but there are some general protocol maybe helpful (see attachment).
If you do IHC-P, you may consider the quality of your antibody. you can also try antibody from another source, here i recommend for you:

If antibody from different source give a same result, you may confirm it.
Thank you Lu for the links, if troubleshooting fails then we will consider changing to a differrent antibody. And that's right, the current antibody is from SantaCruz and the samples are formalin fixed paraffin embedded sections.
