I want to know about calpain1 and calpain2 inhibition via calpastatin in vitro but i want to know more about calcium influx inhibition as a tool for axonal degeneration reversal mechanism?Possible role of above mentioned proteins if anybody knows?
Calcium flux activates pathways ie PLC and also results in calpain activity which may lead to degradation of some of your proteins of interest off the membrane that you have mentioned e.g.NCX3,AnkyrinG,Spectrin. We have recently published a similar function of calpain in the rapid detachment and/or degradation of cell membrane-localized proteins (particularly with lipid-binding domain such as PH domains and polybasic regions. This pattern of Ca-calpain activity can rapidly modify cellular responses that require quick modifications.
