I have 500 ml stock culture which consist of 10^8 bacteria cells. I need to inoculate 10^5, 10^6 and 10^7 cfu/ml to my fish samples. Which way should I follow from now on? Should I harvest the cells with centrifuge or directly dilute from the stock culture?
Dilute your stock culture 100X and 1000X times and then inoculate the bacterial samples
Dear Ms. Agrawal many thanks for your suggest. I also would like to learn that; is it correct to inoculate 1 ml bacteria culture to 10 g fish flesh. or total weight should be 10? I mean 9 g fish 1 ml bacteria culture?
Dear IIknur, you take 10 microlitre of your stock and make it to 1 ml (100X) and similarly take 100 microlitre of stock and make to 1 ml (10X) and then inoculate this 1 ml of diluted sample to 10 gram of fish sample. In first dilution the overall concentration of the bacterial sample inoculated/ gram of fish would be 1000X ( 10^5 bacteria per gram fish) and in the second dilution the overall concentration of the bacterial sample inoculated/ gram of fish would be 100X (10^6 bacteria per gram fish) .
