As I know TSB should be stored in cool and dark. I prepared TSB and stored it under cool and dark conditions. But I must make a stock culture with this broth, so after inoculation of bacteria should I continue to support dark condition? If not the broth will be spoiled?
Well, after inoculation I have incubated TSB on a light room, and the microorganism I have been working, have grow without any problems, so you can try
You can store the broth after sterilization in a refrigerator for 14 days.
In general incubators (except algal incubators) are dark once we close the door. Except few phototrophs you can culture bacteria in dark or with light it doesn't effect the growth. The only thing you have to keep in mind is before you make the stock make sure the bacteria has grown to a sufficient numbers and free from contamination. You can use 15 to 20% glycerol (sterile) to make stock cultures and tore them at -80C.
Trypticase soy broth is not so delicate a medium as you are apprehending it to be. You just continue to work in a normal way after inoculation. If I am not wrong, then most of the time our incubators are dark, so there is no need to have any special dark conditions for ioncubation of your inoculated broth. Of course the dehydrated medium is to be stored under cool and dark condition as otherwise it may get contaminated with other organisms and moisture from air, which will spoil the medium. Sterilised medium can be stored in room temperature (25-28 C), which you must do at least for 24 hrs, to check the efficiency of sterilization.
The cool dark storage is really for long term stability of the medium, like weeks or months. Light for a few days is not going to have any adverse consequences.
Just to follow up, I typically store TSB in the refrigerator for 1-2 weeks and have not had any adverse side effects.
