I will inoculate the bacteria in fish and then I will treat with high pressure. I would like to know if it is correct to put the fish samples after inoculating with bacteria in the fridge (4oC) overnight (or less) to let the bacteria adapt? I need the growing of bacteria to see the effects of high pressure.

I'm quite interested in the outcome of your experiment. what type of bacteria are you inoculating? what is the usual storage and transport condition of the fish? I think it should be refrigerated, but I think you need to mimic the real conditions in order to validate the technology for industrial employment. Please let me know...

How big is the pressurization chamber? What are weight marinade ratio, temperature and pressure range, salt concentration range, pH range, storage term range. .. etc?

If it is important to you and your study to maintain relevant bacterial diversity to that of the industry, you want to store at 4C. However, I've seen these types of bacteria take as long as a week to thrive at 4C; when artificially applied. If I were you, I would just inoculate the bacteria in broth and observe its turbidity over time at 4C.
As a side note, if you are only concerned with bacterial presence (regardless of the bacterial genera), storing a retail purchased fish sample in 4C overnight is enough for sufficient bacterial growth. However, if Photobacterium and Morganella are the bacteria of your concern, you definitely want some preliminary observation as the CFU/mL-viability-level of your inoculate could be an important factor to your study.

If you will threat the fish after inoculation with UHP, be sure that you use different strains of the normal spoilage flora of the fish ... maybe 3 spp. Probably they are all negative. Remember that Grampositive mo usually withstand UHP better than Gramnegatives. Maybe you should test one?
Another possibility (maybe more reliable) is not the inoculate the fish, but to use its natural microflora. Make plate counts of the most important groups of mo. Preferably from 3 fishes before and 3 fishes after UHP. Repeat this experiment at least three times with the same type of fish. Thereafter you have a good idea about the effect of UHP on that type of fish.
