We used by the past murine colon carcinoma CT26.WT (ATCC code CRL-2638) cells, which can be obtained from the American Type Culture Collection (ATCC; Manassas, VA, USA).
Subcutaneous CT26.WT colon cancers are obtained by injecting one million murine CT26.WT cells into the left flanks of female Balb/c mice (Charles River, Arbresle, France). Orthotopic colon cancer models can be obtained by injecting one million CT26.WT cells in the caecum of six-week old female Balb/c mice (23+2 grams; Charles Rivers, France). Orthotopic grafts are performed under anesthesia by an intraperitoneal injection of 112.5 mg/kg body weight (bw) of ketamine (Ketamine 1 000, Ceva, Libourne, France) and 1.5 mg/kg bw of xylazine (Proxylaz, Prodivet pharmaceuticals, Eynatten, Belgium).
