Dear researchers,
I am working on cancer cell and human RBC cells.Testing cell viability,we using different assay like MTT and trypan blue.Now we have spectrofluorimeter.Can anyone tell me how to measure the PI uptake in Cancer cells and human RBC cells(by spectrofluorimeter)

i can't tell you the exact concentration of PI that should be added but it is very simple-
you just add the PI for 10 min or so at RT and then wash excess of PI put in PBS and measure in a suitable wavelength at your fluorimeter. (96 well plate)
just like you do MTT or MTS.
I would use scaling up of PI just to verify the maximum uptake on normal vs treated cells .
