i am working on anticancer drugs that may cause oxidative stress in cancer and normal cells.my interest is, treatment of that drug to RBC cells in invitro and to check the ROS generation on the cells.
We dont have flowcytometer but we have flurocent spectrophoto meter.here,my question is ,the sample preparation for ROS is same like flow cytometer or different.can you please help me in this.
Another option may be measuring the level of isoprostanes generated after the treatment. You can buy the 8-Isoprostane ELISA Kit from cayman chemical (https://www.caymanchem.com/product/516351).
A combo of tests can be done..try the dcf-da assay alongwith catalase & SOD activity (kits) as well as immunoblots for same..then you may further add any of the tests for oxidized GSSG level.
