Hi, colleagues, can I leave a SDS-PAGE overnight after running? Should I leave it in fixation solution (50% methanol, 10% acetic acid) or shall I perform the fixation for 1 h and then leave it in water? Thanks for the help.
Leave it in the fixation solution. Just change it before you leave.
For me. I also leave it in fixation solution but sometimes the band would not be as clear as 1 hour fixation.
The gel can be left in fixation solution and rinsed thoroughly with water before silver staining to avoid background in the gel while developing
Once fixed and washed your gel should be OK to store over night. Otherwise bands or spots would be not be sharp.
