Hi all,

I did an immunofluorescence against c-fos in mouse's neurons using a monoclonal rabbit antibody (Cell Signaling) and a secondary Alexa 488 against rabbit (Invitrogen). I saw labeling in the cytoplasm (I labeled the nucleus with DAPI). I was expecting to see labeling inside the nucleus. Have you ever seen c-fos labeling in cytoplasm (I don't see any labeling inside the nucleus) ?

I perfused the mice 2h after the behavior with 4% PFA.

I did my IF on frozen sections mounted on slides.

Also, I have a basic question, I used to dehydrate my slices before mounting with H2O-Ethanol-Xylène. Is it usefull to do it or not ? My mounting medium is Fluoromount.
