I would like ask whether blood or colon tissue could be a better option to analyze TNF-alpha, IL-1B, IL-10, IL-6 levels in mice colitis model.
Also which method would be a better option btw flow cytometry and ELISA and why?

Blood or tissue: What do you want to look at? The markers in circulation or at the site of the disease? It really depends on your goal. Usually, most people would do both especially, if they are looking at biomarker identification.
Flow or ELISA: Well...ELISA is easier. Flow you will usually need to restimulate and culture with an inhibitor so you can measure the cytokine intracellularly. However, flow will give you more data as in what cells are making what.
You can use both blood (serum) and tissue for cytokine examination as they both would have a significant amount of cytokines. ELISA will be easier than flow.

You can use PBMCs isolated from blood to analyze these cytokines. In 2019, my article published in Q1 journal titled " Immunomodulatory properties of selectively processed prawn protein fractions assessed using human peripheral blood mononuclear cells" can be helpful as reference. Here I have analyzed the level of IL-4, IL-10, IL-17 and IFN-gamma for allergy analysis.
