I am planning to transfect COS-7 cells and it is my first time working with them, so do you have any advice how to optimize the transfection conditions ?
can i use these cells at first passage or only after few passages ?
how many cells to seed in a 6-well plate ?
and how confluent they should be at the time of transfection ?
The conditions for transfecting COS-7 cells are largely like HeLa cells. You can transfect them at first passage, yes. Seed the same quantity as you would for HeLa. They should be about 70-80% confluent at the time of transfection.
What's even more fun, you can actually transfect them at the time of seeding - it works with our COS-7 cells and saves you a couple of days.
