Since you are working with 4T1, may i ask something? I guess you also do transfection, which transfection reagent do you use? I am having trouble with transfecting 4T1 cells..

Hi Elan,

I know its late to reply but if at all it help. 4T1 can be easily transfected through Lipofectamine 3000 following the standard protocol. I have done it many time with different shRNA plasmids with very good transfection efficiency.

Also you will need higher concentration (5-10 ug/ml) of puromycin for selection.
