Hello fellow scientists. I am planning to do an ATAC-seq run in the near future and have a few questions and concerns.
I have acquired the protocol used by a previous member of the lab I am in, which was essentially this protocol: Article ATAC-seq: A Method for Assaying Chromatin Accessibility Genome-Wide
(recommend by Illumina)However the protocols use discontinued products such as the Tn5 transpase and 2X TD buffer.
1) My first question is:
Is the 'llumina Tagment DNA TDE1 Enzyme and Buffer Kit' just their Tn5 transposase and the 2X TD buffer?
2) My second question is: Is there a cheaper alternative? This kit is extremely expensive.
3) My Third question is: Is anyone familiar with this protocol? if I use the illumina enzyme do i need to use the illumina kit?
4) My fourth question is: the protocols all refer to "custom illumina primers". I dont know what this means! None of the protocols describe designing custom primers! Can anyone offer insight?
