I collected my samples in RLT and stored at -80c. Later on, i am considering to use TRIzol extraction instead of Qiagen's protocol. The question is: may I add TRIzol to the sample and perform the TRIzol/clorophorm classical protocol? Or, by the contrary, all the stuff in the RLT buffer may affect the efectivity of TRIzol protocol?

Hi Hector! Adding RLT lysis step of course may affect the effectivity of TRIzol protocol. However, RLT buffer contains a high concentration of guanidine salt, which is also used in TRIzol (a monophasic solution of phenol and guanidinium isothiocyanate) extraction. My impression is RLT buffer won't affect extraction much at all. Good luck with your work!
