I’m currently attempting to dissolve ISX-9 for induction experiments but encountered precipitation issues. Here’s my protocol and the problem:

  • Initial Dissolution: Dissolved 50 µg ISX-9 in 500 µL DMSO (stock concentration: 100 µg/mL). Added 50 µL Tween-80 to improve solubility.
  • Dilution Issue: Upon further dilution in either PBS or saline, the solution turned milky/opaque, indicating precipitation.
  • Desired Working Concentration: Target: 4.27 mM working solution.
  • Key Questions:

    • How can I improve solubility to avoid precipitation? (e.g., alternative solvents, sonication, heating?)
    • Storage Conditions: Should the working solution be stored at 4°C or -20°C? Is aliquotting recommended to avoid freeze-thaw degradation?
