I need to prepare a crude extract of Penicillium herguei to evaluate the activity of an intracellular enzyme. Do you think maceration is a good technique?
For enzyme purification, we normally we grind the mycelium under liquid nitrogen and afterwards extract the mycelial fragments by shaking in the desired buffer. You could also consider homogenisation of mycelium in a mixer or in an Ultraturrax device, but grinding under nitrogen proved to cause less activity losses (please use a fume hood for all these procedures that necessarily produce aerosols).
