My lab is interested in extracting RNA from sperm (small RNAs specifically), and I believe we have the correct kit (Zymo Quick-RNA Micro-prep), but we already have a lot of trouble with extracting DNA/RNA from other tissue, and I get the feeling that sperm may even more finicky due to low RNA content. We're also not entirely sure about our sperm isolation protocol, for which we remove epididymis from a recently euthanized mouse. Does anyone have any protocols for this procedure specifically? And do you have any tips for DNA/RNA extraction from sperm? Thanks!
