Use Qiagen FFPE tissue kit or equivalent, but first treat the paraffin sections with tolyene (2 times) and rehidrate with 96% ethanol, followed by 70% and 50%, dry down, and start isolation with the kit.
thank you for your answer, i have already used it (Qiagen)but it takes a lot of time to get it from commercial so i'm looking for classical methode (proteinase K , lysis Buffer ).
From my experience the best method for small FFPE biopsy samples resulting in best quality was using the RecoverAll™ Total Nucleic Acid Isolation Kit for FFPE now from Thermo, previously from Life/Ambion (https://www.thermofisher.com/order/catalog/product/AM1975)
You can do TriZol if you don't want to use a commercial kit or in-house phenol-chloroform extraction.
