There are many primers available in paper published, but most of all have problems with Tm, product size, nonspecific binding and many more. I need a best primer sequences for TB PCR. Is there anyone who has primers and using for TB complex PCR beside any problem?

We have used primers from IS6110 sequence for PCR without any hindrance. You can follow the same PCR conditions that we used (Pubmed IDs: 20644311, 20799764, 20966559, and 21738331). Good Luck!!

You can use oligonucleotide primers within the IS6110 insertion sequence, designated primers Pt8 (5'-GTGCGGATGGTCGCAGAGAT-3') and Pt9 (5'CTCG
ATGCCCTCACGGTTCA-3'), which generate an amplicon of 541-bp. This is one of the widely used primers, and we have also used it satisfactorily for our experiments.
