Generally, kit manufacturer provide full details about the kind of samples that could be tested. Alternatively, I will suggest you to prepare homogenates adjusted to be 1 or 2mg protein per ml of lung or any other tissue homogenate from a control organ just to have an idea before going to actual experiment. From this you can take 100 ul or 50 ul tissue homogenate for lpo experiment and measurement purposes in a total reaction volume of 2 or 3 ml.

I think i can measure total protein concentration with Bradford's method. I will try. Thank you for your help.

I've never used the MDA Elisa Kit (E-EL-0060), usually I use the thiobarbituric acid reactive substances (TBARS) protocol (see chiyama and Mihara, 1978). And so I use phosphate saline buffer to prepare my samples, according to sample weight (ex.: 150 mg tissue to 300 mL PBS)
