In primary culture you have kind of "developing neurons" which can be detected with doublecortin. There is a Santa Cruz goat antibody (sc-8066) which should be diluted 1:400 - this should detect these neurons!
If you dont have any MAP2 positive neurons then i would include somepoaitive control for the antibody and the method in the first place. Also its senstive to hypoxia, so depending on the culture condition, MAP2 could be affected.
Thanks - I see MAP2 positivity in general with neurons, but there appears to be certain cells that are not positive for MAP2 and are also not glial cells...
