I'm trying to stain the inflammatory cells or immune cells (both or either is okay) from in vivo tissue sections. I implanted the polymeric scaffolds (polycaprolactone, PCL) into rats.
From beginning, I have tried to stain the cells with IHC, but now I'm finding different ways due to too much non-specific binding of antibodies to the polymeric scaffolds I implanted.
Are there any ways to stain the inflammatory cells or immune cells by not IHC (not involving antibodies) but by chemicals?

Neutrophils, granulocytes are positive with PAS reaction.
If you just want to stain all lymphocytes you can do a Giemsa-stain or a HE-stain as overview-stain. Lymphocytes are identified by their typical shape, size, stainability.
In H&E stained tissues, resident macrophages, neutrophils (distinct nuclei), eosinophils (cytoplasmic eosinophilic granules), and general lymphocytes (including plasma cells) are differentiable under light microscopy. Mast cells can be differentiated by Toluidine staining from other inflammatory cells. You can get some advices or helps from some specialists, such as small animal pathologists, to morphologically differentiate the cell types.
