Hello ,
I am having trouble in differentiating U937 cells. I tried using various incubation periods and different PMA dosages—10, 20, and 100 ng—but it seems like many of the cells are dying regardless of PMA concentration. I would sincerely appreciate your advice in this regard if there is anything I should be aware for this experiment.

In our lab, we use instead of PMA a concentration of 50 to 100 ng/mL of M-CSF which is non-toxic.
