Hello,

Do anybody face issues or have concerns about the gRNA scaffold's secondary structure formation. During in-vitro transcription, there is no heating and cooling step to enable the formation of the secondary structure for this scaffold needed to recruit/ form complex with cas9 endonuclease. 

Please let me know if anyone knows any steps we ought to take or if anyone has same concerns.
