My lab has recently acquired a vibratome, and we've been looking for an antifreeze (cryo solution) to store brain tissue at freezing temperature without the liquid freezing.

Mouse/dog brain tissue
40-80microns in thickness
Stored in well plates
Vibratome

We've had excellent preservation of antigenicity of FD sections storage solution from FD Neurotechnologies. The sections are stored at -20 until needed.

we are using: 109621 Ethylenglycol for Analyse EMSURE® Reag. Ph Eur,Reag. USP from Merk
I used below cryopreservative for 40U thick brain sections stored at -60C for more than 2 years without any changes in immunohistochemical stainings.
CRYOPROTECTANT SOLUTION
The solution contains
20% Glycerol
2% DMSO (Dimethylsulfoxide)
Prepared with buffered paraformaldehyde fix or plain buffer (for some special cases where enzyme activity should not be compromised).
Once the barin slices are in antifreeze solution, Can be stored at -40ºC or -20ºC must only be?
