i dont understand your question completely, but the Disc diffusion method means your test organism is spreaded over the agar plate and antibiotic discs in various concentration should be placed over it after incubation period the zone of inhibition is measured and this is called as antibiotic sensitivity this method is called "Kirby Bauer Disc Diffusion method" similarly another method also used that is called agar well diffusion method, for better understanding of agar disc diffusion method please refer the potograph
