I have produced lipase using ssf using aspergillus niger mtcc 872. Now, I want to purify the lipase. During the ammonium sulphate precipitation, it is mentioned 0-80% or 10-80% saturation. what does it mean?

Case:A: If I want to precipitate the protein from 100mL crude sample, do I need to add 10% to first 100mL sample and extract the precipitate

and add 20% ammonium sulphate to another fresh 100mL sample

and add 30% ammonium sulphate to another fresh 100mL sample until it reaches 80% saturation

OR

Case:B: first add 10% ammonium sulphate and then get the precipitate and use the remaining sample and add 10% more to make it 20% saturation and so on?
