We have some backlogged wastewater samples which we would like to analyze for SARS-CoV-2. We are having success with fresh and recently frozen samples (-20 C). However, our oldest samples (May-August) were frozen in larger containers (~ 1L) rather than small aliquots and all attempts to measure RNA via qPCR have essentially come out null (non-detects). The older small aliquots (45 mL) are better but still usually a bit lower in value than expected with a few more NDs then expected. Unfortunately, these samples are just raw wastewater and no preservatives have been added. Is there any way to thaw them to greatly reduce the RNA degradation when doing so? It may be a lost cause but we'd like to see if we can recover something from these samples. Thanks.
