Can you implant an optic fiber for transient illumination without having to leave the cannula implanted? We're trying to setup a photothrombotic stroke model in the mouse hippocampus and need to transiently illuminate the area (10-20 mins) to create the clot and induce the ischemic injury. Since this is a one time thing we don't need to leave the cannula/optic fiber connector implanted.
Is this possible?

I think it is possible. I know the lab of Prof. Bai'Ou Guan @Jinan University (OSA fellow) is doing such kind of things. Please check their paper. And there should be also other lab doing such things as well.

I'm interested in trying this as well (photothrombotic stroke through an optical fiber). In my case I have the fiber implanted already for unrelated reasons but was wondering if you've tried it since your post. And if you'd share the parameters you're using in your approach, i.e. what light source do you use and at what power?

Hi Alice, sorry just saw your post. We tried a few animals with variable success. We're using a 520 nm diode laser from Thor Labs set at 10 mW power. We inject the Rose bengal retro-orbitally 5 min before the stroke and illuminate the brain for 10 mins. We definitely get a stroke, but our main problem is location and size. Most of our strokes are too big/severe and the animals barely survive 24h. We're currently testing different parameters (power and time). Also we do not leave the fiber implanted and re-use it between animals. Shams recovery fine, so I don't think our issue is with the surgery itself, but rather with the stroke parameters.
