Any pointers on how to get Cryptosporidium sporozoites to glide on non-cell substrates? We've tried oocysts from a couple of different sources (WaterBourne and the production lab at U of Arizona, winter harvest), different substrates (plastic, glass, PLL, protein), different media (culture medium, HBSS, low calcium, and homemade Ringers from David Sibley's recipe), room temperature and 37C, and low light (observed by intensified CCD at limiting illumination). We have yet to capture a single gliding cell. Any and all ideas and trade secrets appreciated!
