I have a problem with absolute quantification and standard deviations.

When I do my standard curve (with gBlocks) and run all my samples the standard deviation between triplicates are around 0,1 or 0,2 which is still acceptable.  But when I calculate the standard deviation for absolute quantification (absolute number of molecules) the deviations are around 10% of molecules.

I was looking around for articles that would  present this question in the way that can be used for publishing, but had no luck. 

If any of you have any idea how to deal with this problem, it would be helpful. And If any of you have any articles that represents the methodology and statistical analysis of absolute quantification with alternative splicing, I would really appreciate.
