DNA was amplified using a PCR mix that included SYBR green dye. Could this DNA amplification undergo pyro sequencing without interference from the dye?
i would think that in pyrosequencing the pcr product is labelled with biotin on one strand which is bound to a streptavidin column and the unlabelled strand is denatured off to leave single stranded DNA so releasing almost all of the sybr green. The sequencing reaction is then carried out on the single stranded dna so I would expect it to work fine
