I have used different recipe of physiological buffer solutions for live cell imaging previously (such as live cell imaging solution from Thermofisher). Based on Thermofisher's description, " It is an optically clear, physiological solution buffered with HEPES at pH 7.4 that keeps cells healthy for up to 4 hours at ambient atmosphere and temperature. "
Recently we installed a chamber to our microscope to maintain temperature (at 37degree) and CO2 level, and wondering if I can still use those physiological buffer solution instead of complete media. Do you expect cells to respond differently at 37 degree when they are in the physiological solutions?
