I am culturing C2C12 myotubes for immunofluorescence on collagen-coated plates and wondered if I can sterilize the plates after the collagen is dry with UV light? Will it damage the collagen, so it will become less effective in promoting cell attachement to the plate? Has anyone tried that?
Thanks! We do not use precoated plates and coat them by ourselves. Usually I can store them for a couple of days before seeding the cells. This is why I wanted to sterilize the plates.
No. It will neither affect the collagen nor the attachment of the C2C12 cells. However, C2C12 cells grow well in normal cell culture dishes, and glass coverslips without collagen coating .

Most collagen sellers as Sigma and others describe UV as sterilization method for manually coated plastics and coverslips. I extracted collagen from rat tail and i coated tissue culture dishes and coverslips, then i exposed them to UV up to 2 hours under the hood and all give me good attachment and spreading. Finally, i think UV can affect collagen during the process of cell formation of collagen but not the mature collagen.

I would suggest not to use UV light, hence it would probably denaturate collagen.
I've used UV sterilization for collagen plates using rat tail collagen I and human placenta Collagen IV. It works just fine and my kidney cell lines attach an activate just like when I use commercially coated plates. The only downside is with longer UV exposures it will make the plastic less transparent. The Sigma protocol online works great for me.
When I steriled the MPC-BMA copolymer coated plate with Gamma ray and UV light,the gamma ray breaking the coating film and cell adherence while UV light treatment cell can form spheroid body and not adherent on the plate surface. So I think using UV light treatment the coating plate is more safe than Gamma ray treatment.
