Addition of both LPS and IFNg stimulates the macrophages. Upon infection and stimulation of macrophages with LPS and IFNg, will the cells get stressed that one could not observe the infection of cells by virus in the first place?
How good are RAW 264.7 culture in phagocytosis upon infection?
Stimulation of RAW 264.7 macrophages with LPS (10 ng/ml) enhance its potential of phagocytosis as well as proliferation of the cells. We did the phagocytosis assay in our experiment previously with LPS and some other compounds and found that all the sequential steps of phagocytosis like membrane fluidity, phagolysosome fusion etc. were enhanced in RAW 264.7 cells. Well I'm not sure about IFN gama but you can use LPS for your experiment.
Thank you. Did you carry out the assay with virus infected cells? and also i am trying to get hold of the assay using Vybrant phagocytosis assay kit. Can you suggest other protocol to carry out the same?
