Hello,everyone.I want to know why we use Cs internal solution replace K internal solution ,when we want to record EPSC ,IPSC or AMPA NMDA ratio? If I want use K internal soluton to record AMPA NMDA ratio,It can work? I also read some discussion ,they say good space clamp when using Cs, but I want to know whether K is ok and the result if use K?

Hi Guangling!
If using physiological (1mM) levels of Mg+ in your aCSF, you would need to record NMDAR-EPSCs at +40mV to remove the Mg+ block. To get there, you probably need to use Cs-based intracellular solution for better space clamp.
An alternate approach is to clamp at -70 mV, use low (0.1 mM) Mg+, and DNQX/D-AP5 to obtain the AMPAR/NMDAR ratio.
