I am facing a problem during transformation. Whenever I use LB agar plates with either kanamycin or amp my plates get dried overnight. Sometime I see colony like dots on plates sometime clear but dried agar. I usually prepare agar and store it. Whenever required we melt it and pour on plates. However some times i do get normal results for same batch plates. Has anyone faced same problem? Please help me in solving it.
It is possible the plates are poured with too thin a layer of agar. Another option would be to incubate the plated in a chamber with some moist paper towels to provide a little humidity (like in a zip bag). Or use shrink wrap to seal the agar plate. It sounds like your main problem is rapid loss of moisture.

I agree with Widen, this problem cause of your thin layer of agar. Also pleaase check your incubator humidity. good luck for you
I think you should pour the plates after autoclaving and then plates can be stored in low temperature.Might heating of the agar leads to the reduction in moisture.

Does this happen when you dry plates taken out from 4C? If so, it is due to the immediate difference in the temperature. I get this sort of bubbles (appears like a colony) when I dry plates at 37C or 28C taken out from 4C. Leaving the plates in room temperature for 3-4 hrs prior to drying at 37C will solve this issue!
