Before cryosection, tissues can be simply fixed (4% Paraformaldehyde and then sucrose dehydration). In theory, any tissue fixed by paraformaldehyde should go through an antigen-retrieval procedure, because the paraformaldehyde can cross-link the protein. However, when I read some papers dealing with cryosection, the steps are always: fixation---cryosection---IF/IHC, the antigen-retrieval step is always skipped. Can anyone help me figure it out?
