I am working in a laboratory with Cov19. Extraction is doing kit but it isn't good and the amount RNA, 260/280 is higher than 3. Can someone help us in this regard?
"Abnormal 260/280 ratios usually indicate that a sample is contaminated by residual phenol, guanidine, or other reagent used in the extraction protocol, in which case the ratio is normally low." Do you use Trizol based extraction method?
A260/280 ratio greater than 2.2 indicates the presence of basic contamination whereas if ratio decreases 1.9 indicates the presence of acidic phenol or proteins.
Just use TRIzol and Chloroform then wash it with Isopropanol and then Ethanol. Be sure when taking the aqueous phase after centrifugation, you're not taking other layers.
