Mouse intestinal organoids were growing fine until 3 passages. After that, I passages them last Friday and left for the weekend. Monday I found that they were dying. I am not sure what is the reason for that. I am using the protocol of Intestinal Epithelial Organoid Culture with IntestiCult™ Organoid Growth Medium (Mouse). Instead of matrigel dome , I am using Cultrex UltiMatrix Reduced Growth Factor Basement Membrane Extract dome. Any suggestions would be greatly appreciated.

I'm not an expert in intestinal organoids, and its hard to see in the pictures, but could there be a contamination?Or could the media components be expired or stored wrong? Usually they are only good for 2 weeks in the fridge.

It looks like the culture was contaminated between Friday and Monday, when the images show dead organoids and a significant increase in the amount of debris in the dome. I suspect the buildup of dark patches in the dome is from the contaminating organism expanding in the culture.
