I conducted mdamb 231 cells scratch assay in 6-well plate. Tried taking images under microscope (10X) for 0th hour right after the assay, but couldn't able to take properly. Lookslike some of the cells have detached as whole mat or stressed or appeared blur. Is it beacuse of the confluency?

Yes, it sounds like the cells were a detached confluent sheet. Do you have a picture before making the scratch?

No I haven't taken images before the assay. But the wells were confluent. I have attached the image taken at 0th hour

When the scratch was made, was there medium in the well or were they exposed to air? Could they have gotten too dry?
